Manuscript proposed by Yihao and co-workers (applsci-1942164) entitled “Trypsin inhibitor from soybean whey wastewater: Isolation, purification, and stability” presents the soybean trypsin inhibitor isolation from simulated soybean whey wastewater through a sustainable method. Additionally, the effect of temperature, pH, and pepsin on the STI stability was discussed. In my opinion, the manuscript presented deals with very basic aspects of scientific research, the experimental procedure is underdeveloped and does not allow drawing conclusions, additional experiments are needed.
My major comments are presented below.
Major concerns:
- what is the novelty of presented methodology and presented topic?
- the Authors presented the utility of the proposed method however, in my opinion the correctness of the actions taken has not been proved in the text,
- Introduction – more details about the methods used in the analysis of STI in soybean whey or soybean whey wastewater should be presented
- Result section/discussion – in my opinion, additional experiment where STI is investigated in soybean whey wastewater should be presented. It was suggested in the title. Therefore, in my opinion the content of the work is not related to the topic specified in the title
– how was the nature of separated/isolated compounds determined? How the nature of isolated protein was confirmed?
- figure 8 – H2O – use subscript; error bars should be presented
- figs 6 and 7 – the presented results are interested in particular that the optimal pH for STI is 8.0 and the temperature 35C.
- statistics analysis is limited

Title: Trypsin inhibitor from soybean whey wastewater: Isolation, purification, and stability
The study aimed to design a simple method for isolation and purification of STI from soybean whey.
The research has set up well, the writing is good, but some corrections are needed before the publication.
Comments:
The key words should provide in alphabetic order and each word should be capitalized.
Line 25: replace "and" with ","
Line 28: are you sure? "twenty tons of soybean whey are generated per ton of soybean protein isolate "??? How is it possible? 20 tones per ton?
Line 58-64, Line 95-105 and Line 116-121 add the references.
Line 107 H2SO4 has to change to "H2SO4"
Change all " r/min " into " rpm "
Line 126 state the concentration of standards.
Are Line 80 "BAPA" and line 138 "BAPNA" the same?
Add some detail to material and methods: Section 2.4.5.1 (state the pH of samples), Section 2.4.5.2 (state the temperature of conditions), Section 2.4.5.3 (state the pH of samples and the temperature of conditions), section 2.4. (Add the mean comparison test name).
Line 194 "and4 " add a space
Line 224, 252, 286, 308, 325, 337, 372 state the name of statistical test
In the Figure 2: I think the letter "bc" denotes for recovery rate of soybean whey protein under pH of 7.5 is not correct. It should be "b", please re-check the statistical analysis result.
Figure 3, Figure 6-a, Figure 7-a : Y axis had to start from zero
Line 271-273 state the exact conditions which the KTI (1733.5 TIU/mg) and BBI (2588.3 TIU/mg) were obtained.
Figure 5 the letters on the columns seems wrong. As a rule of thumb the letter "a" denotes the greatest value. So the letter "a" seems to insert for the two columns on the right. Re-check please.
Table 1- the letters of significance are not provided.
Table 2 the significance letter for "β-turn" and" Random coil" are not correct. Re-check them.
Line 387-391 it is better to move "It can be used as a nano carrier material for encapsulating insulin, functional short peptides to avoid hydrolysis in the gastrointestinal tract and improve bioavailability. The successful extraction of high value-added soybean trypsin inhibitor from soybean whey wastewater provides an idea for the high value utilization of soybean whey 390 wastewater." to introduction.
