Cover Story: Scorpionates, tridentate ligands known for their versatile coordination properties, have emerged as promising scaffolds in cancer therapy. This review explores their unique potential, focusing on metal-based complexes like copper and ruthenium, which exhibit cytotoxic activity by disrupting mitochondrial function and generating reactive oxygen species. Ligand modifications, such as sulfur heterocycles and unsubstituted pyrazoles, enhance stability and selectivity. Despite significant findings on these ligands, scorpionates remain underexplored in systematic structure–activity studies. Their diverse chemistry and selective biological mechanisms hold potential for next-generation anticancer therapeutics. View this paper
