Cover Story: Cell-free DNA (cfDNA) is fragmented extracellular DNA that is released from cells into various body fluids. Previously published data supports cfDNA as a biomarker in adult horses, but shows that the plasma matrix effect interferes with direct measurement. This study also found evidence for the matrix effect when directly measuring cfDNA in foal plasma. However, even with extraction, there was no significant difference in plasma cfDNA, or cfDNA/neutrophil ratios, between healthy, SNS, or septic foals. In future studies, investigators should be aware of the matrix effect when measuring plasma cfDNA in foals. Further research is needed to determine the capacity of foal neutrophils to perform NETosis, and whether serial measurement, or measurement after plasma replacement therapy, impacts cfDNA as a biomarker for sepsis in foals.
