Cover Story: Cutaneous T-cell lymphoma (CTCL) is a rare form of non-Hodgkin lymphoma marked by the malignant expansion of T cells in the skin. Despite treatment advances, its cure remains elusive. The dysregulation of the JAK/STAT pathway plays a central role in CTCL pathogenesis, making it a promising target for therapeutic intervention. Initially approved for inflammatory diseases, JAK inhibitors are now being explored for CTCL. This review highlights clinical and preclinical studies on six JAK inhibitors and various therapeutic approaches, as well as cases of CTCL relapse following JAK inhibitor therapy. While these findings suggest the promise of targeting the JAK/STAT pathway in CTCL, further research is needed to assess long-term safety, reduce adverse effects, and optimize treatment strategies. View this paper
