Technologies, such as Chat Generative Pre-Trained Transformer (ChatGPT, Smart PLS version 4), are prime examples of Generative Artificial Intelligence (AI), which is a constantly evolving area. SMEs, particularly startups, can obtain a competitive edge, innovate their business models, gain business value, and undergo a digital transformation by implementing these technologies. Continuous but gradual experimentation with these technologies is the foundation for their adoption. The experience that comes from trying new technologies can help entrepreneurs adopt new technologies more strategically and experiment more with them. The urgent need for an in-depth investigation is highlighted by the paucity of previous research on ChatGPT uptake in the startup context, particularly from an entrepreneurial perspective. The objective of this research study is to empirically validate the Generative AI technology adoption model to establish the direction and strength of the correlations among the adoption factors from the perspectives of the entrepreneurs. The data are collected from 482 entrepreneurs who exhibit great diversity in their genders, the countries in which their startups are located, the industries their startups serve, their age, their educational levels, their work experience as entrepreneurs, and the length of time the startups have been on the market. Collected data are analyzed using the Partial Least Squares Structural Equation Modeling (PLS-SEM) technique, which results in a statistical examination of the relationships between the adoption model’s factors. The results indicate that social influence, domain experience, technology familiarity, system quality, training and support, interaction convenience, and anthropomorphism are the factors that impact the pre-perception and perception phase of adoption. These factors motivate entrepreneurs to experiment more with the technology, thereby building perceptions of its usefulness, perceived ease of use, and perceived enjoyment, three factors that in turn affect emotions toward the technology and, finally, switching intentions. Control variables like age, gender, and educational attainment have no appreciable effect on switching intentions to alternatives of the Generative AI technology. Rather, the experience factor of running businesses shows itself to be a crucial one. The results have practical implications for entrepreneurs and other innovation ecosystem actors, including, for instance, technology providers, libraries, and policymakers. This research study enriches the Generative AI technology acceptance theory and extends the existing literature by introducing new adoption variables and stages specific to entrepreneurship.

The present study evaluated the applicability of 1H NMR and UV-Vis spectroscopies as analytical techniques for the characterization and determination of biodiesel conversion and for monitoring the oxidative stability of biodiesel samples with antioxidants. For this study, safflower and babassu biodiesels were obtained through transesterification, and physicochemical properties confirmed the success of both reactions. A bench-top accelerated oxidation system was used as an alternative to the Rancimat® method, with samples of 6.0 g heated at 110 ± 5 °C and collected every 2 h for 12 h. The agreement for biodiesel conversions was good, with divergences between 2% and 0.4% for safflower biodiesel and 1.9% for babassu biodiesel. As for UV-Vis spectroscopy, the technique showed the same trend as the Rancimat® method, showing efficiency in evaluating the oxidative stability of safflower biodiesel and in the performance of antioxidants BHT and DMP-30. The accuracy of NMR signals integration for mixtures of safflower oil and safflower biodiesel and the use of UV-Vis spectroscopy associated with a bench-top accelerated oxidation system to investigate the performance of phenolic and amine antioxidants in safflower and babassu biodiesel were explored for the first time, showing results close to the standard methods. Therefore, 1H NMR and UV-Vis spectroscopies could be applied as alternatives to the GC and Rancimat® methods to determine conversion and monitor the oxidative stability of biodiesel rapidly and practically.

This paper addresses the optimization of contracted electricity demand (CD) for commercial and industrial entities, focusing on cost reduction within the Brazilian time-of-use electricity tariff scheme. Leveraging genetic algorithms (GAs), this study proposes a practical approach to determining the optimal CD profile, considering the complex dynamics of energy demand on a city-like load. The methodology is applied to a case study at the Federal University of Pará, Brazil, where energy efficiency and demand response initiatives as well as renewable energy projects are underway. The findings highlight the significance of tailored demand management strategies in achieving energy-related cost reduction for large-scale consumers, with implications for economic efficiency in energy consumption.

This study investigated the processing of lexical elements of idioms in isolation. Using visual word priming, spreading activation for idiomatically related word pairs (e.g., pop–question) was compared to that for semantically related (e.g., answer–question) and unrelated word pairs (e.g., trim–question) in two experiments varying in SOA (500 ms and 350 ms). In line with hybrid theories of idiom representation and processing, facilitatory priming was found in both experiments for idiomatic primes, suggesting a tight link between the words of an idiom that is mediated by a common idiom representation. While idiomatic priming was stable across SOAs, semantic priming was stronger for the short SOA, implying fast and early activation. In conclusion, one lexical element of an idiom can facilitate the processing of another, even if the elements are not presented within a phrasal context (i.e., within an idiom), and without the words being semantically related. We discuss our findings in light of theories about idiom processing, as well as current findings in the field of semantic priming.

Opioids are commonly prescribed for the treatment of chronic pain. Approximately 50% of adults who are prescribed opioids for pain co-use cannabis with their opioid treatment. Morphine is primarily metabolized by UDP-glucuronosyltransferase (UGT) 2B7 to an inactive metabolite, morphine-3-glucuronide (M3G), and an active metabolite, morphine-6-glucuronide (M6G). Previous studies have shown that major cannabis constituents including Δ9-tetrahydrocannabinol (THC) and cannabidiol (CBD) inhibit major UGT enzymes. To examine whether cannabinoids or their major metabolites inhibit morphine glucuronidation by UGT2B7, in vitro assays and mechanistic static modeling were performed with these cannabinoids and their major metabolites including 11-hydroxy-Δ9-tetrahydrocannabinol (11-OH-THC), 11-nor-9-carboxy-Δ9-tetrahydrocannabinol (11-COOH-THC), 7-hydroxy-cannabidiol (7-OH-CBD), and 7-carboxy-cannabidiol (7-COOH-CBD). In vitro assays with rUGT-overexpressing microsomes and human liver microsomes showed that THC and CBD and their metabolites inhibited UGT2B7-mediated morphine metabolism, with CBD and THC exhibiting the most potent Ki,u values (0.16 µM and 0.37 µM, respectively). Only 7-COOH-CBD exhibited no inhibitory activity against UGT2B7-mediated morphine metabolism. Static mechanistic modeling predicted an in vivo drug–drug interaction between morphine and THC after inhaled cannabis, and between THC, CBD, and 7-OH-CBD after oral consumption of cannabis. These data suggest that the co-use of these agents may lead to adverse drug events in humans.

Islam was an important factor in the decolonisation of Muslim countries from European colonial rule during the 19th and 20th centuries. However, Muslims are among the migrant-settler populations of Australia, Canada, the United States, and other British colonial states that continue to dispossess and disenfranchise Indigenous populations. This article contributes to the debate on “decolonising Islam”. It contends that covenants with God and between people in Islam’s pre-eminent sources, the Qur’an and sunnah, are antithetical to colonialism and reinforce a praxis-orientated decolonial–Islamic agenda. This article focuses on three aspects of decolonisation, addressing: (1) supremacist ideology; (2) human existence and coexistence; and (3) claims of entitlement. Using Australia as the primary case study, it examines Islamic obligations towards Indigenous peoples in settler-colonial states, emphasising the potential of covenants to promote mutual recognition and dialogue towards redressing injustices and building respectful coexistence.

Sildenafil is used to treat pulmonary hypertension in neonatal intensive care unit (NICU) settings. As multiple intravenous (IV) medications are co-administered in NICU settings, we sought to investigate the physicochemical compatibility of sildenafil with a range of IV drugs. Sildenafil 600 mcg/mL or 60 mcg/mL was mixed 1:1 with the secondary drug solution to simulate Y-site co-administration procedures. Physical compatibility was evaluated by visual observation against a black and white background and under polarized light for two hours for changes in colour, precipitation, haze and evolution of gas. Chemical compatibility was determined from sildenafil concentrations, using a validated, stability-indicating high-performance liquid chromatography assay. Sildenafil 600 mcg/mL was physicochemically compatible with 29 of the 45 drugs tested at ‘high-end’ clinical concentrations and physically incompatible with 16 drugs and six ‘2-in-1’ parenteral nutrition solutions. Sildenafil 600 mcg/mL was compatible with lower, clinically relevant concentrations of calcium gluconate, heparin and hydrocortisone. Aciclovir, amoxicillin, ampicillin, ibuprofen lysine, indometacin, phenobarbitone and rifampicin were incompatible with sildenafil 600 mcg/mL, however these IV medications were compatible with sildenafil 60 mcg/mL. Sildenafil 600 mcg/mL and 60 mcg/mL were incompatible with amphotericin, flucloxacillin, furosemide, ibuprofen, meropenem and sodium bicarbonate. Sildenafil compatibility with commonly used syringe filters was also investigated. Sildenafil solution was compatible with nylon syringe filters, however, absorption/adsorption loss occurred with polyethersulfone and cellulose ester filters.

Carentonosaurus soaresi was recently described in the uppermost middle Cenomanian (Upper Cretaceous) of Casais dos Carecos (Coimbra, western Portugal) based on a diverse set of new material (cervical and dorsal vertebrae) of the Pythonomorpha lizard. The main morphological characteristics observed in the vertebrae used for the diagnosis of this species are the presence of distinct lateral and subcentral foramina, highly laterally projected paradiapophyses beyond the prezygapophyses, a low subrectangular neural spine ornamented with longitudinal grooves, and dorsal vertebrae displaying a sagittal furrow along the ventral surface. Additional diagnostic details observed both in new material and the previously studied vertebrae are described herein, such as the presence of keels in the zygantrum and zygosphene. These and other important morphological characteristics present in the species soaresi are absent in the genotype Carentonosaurus mineaui and in other known Squamata, allowing for the definition of the new genus Segurasaurus.

Pericarp colors are critical agronomic traits that affect the quality and economic values of fruits. Although a diversity of bitter melon pericarp (BMP) colors is available, the fruit pigmentation mechanisms remain elusive. Hence, this study aimed to unveil the key metabolites and molecular mechanisms underlying variation in BMP coloration through integrative metabolomics and transcriptomics analyses of four differently colored genotypes, including K1102 (grayish orange), 262 (grayish yellow), 1392 (very soft green), and K115 (dark grayish cyan). The four BMPs exhibited significant metabolite profile and transcriptional differences, as over 112 and 1865 DAMs (differentially accumulated metabolites) and DEGs (differentially expressed genes), respectively, were identified. The variation in the content of six anthocyanins, including malvidin 3-O-glucoside, petunidin 3-O-glucoside, rosinidin O-hexoside, cyanidin, cyanidin 3-p-hydroxybenzoylsophoroside-5-glucoside, and pelargonidin 3-O-beta-D-glucoside, might be the major driving factor of BMP color changes. Notably, malvidin 3-O-glucoside, rosinidin O-hexoside, and petunidin 3-O-glucoside are the dominant pigments in K115, while carotenoids and other flavonoids may contribute to other colors. Candidate flavonoid structural and regulatory (MYBs, NACs, MADSs, bHLHs, and bZIPs) genes were identified. Of them, gene13201 (anthocyanin reductase), gene8173 (polyphenol oxidase), gene2136 (NAC43), gene19593 (NAC104), and gene15171 (tetrapyrrole-binding protein) might play essential roles in K115 pericarp color development. Our findings deepen our understanding of BMP pigmentation and provide fundamental resources for higher-valued bitter melon breeding perspectives.

Background and Objectives: This study examined the influence of stationary bikes and elliptical machines on knee movement and joint load during exercise. Materials and Methods: Twelve healthy male participants engaged in pedaling exercises on stationary bikes and elliptical machines at speeds of 50 and 70 revolutions per minute (rpm). Knee movement and joint load were assessed using a motion analysis system. Results: The results indicated that elliptical machines induced higher knee joint torque compared to stationary bikes. Notably, peak torque occurred at different joint angles, with stationary bikes reaching an earlier peak at 70°–110° and elliptical machines showing a later peak at 135°–180°. Increased pedaling speed correlated with higher peak knee joint torque on both machines. With the elliptical machine, a higher pedaling frequency correlated with increased peak forces on the knee and ankle joints, as well as vertically. Interestingly, both types of equipment were associated with enhanced peak knee joint torques during high-speed pedaling. Conversely, constant pedaling on elliptical machines limited the ankle angle and could induce inward rotation. Conclusions: This study focused on knee joint torque variations during pedaling on indoor stationary bicycles and elliptical machines. Elliptical machines showed higher peak values of forces and torque, particularly during the propulsive and recovery phases, indicating potential challenges to the knee joint. Notably, peak pedal angles occurred earlier on indoor stationary bicycles, emphasizing the impact of equipment choice on joint kinetics.

Dry lubricants used in highly loaded rolling bearings are in the focus of current research. In previous studies, graphene platelets applied as dry lubricants on the surfaces of angular contact ball bearings demonstrated superior properties. These specific bearings, experiencing both rolling and spinning motion, create more severe conditions for dry lubricants. To gain deeper insights into the lubrication effects, micro-tribological studies were carried out on the respective film formation and running behavior effects. In the tests, a fixed steel ball slid against an oscillating counterpart under a defined load. During the measurements, the applied load and tangential forces on the ball were recorded to calculate the friction. Comparative investigations included nano-graphite particles and fullerene as dry lubricants, in addition to graphene platelets of various staple thicknesses. To increase the adhesion of the films to the surfaces, a pre-rolling process was implemented. Afterwards, the friction on the compressed films was measured. The results indicate that the pre-rolling process effectively reduces the friction of the system. After testing, the surfaces underwent analysis using laser scanning microscopy to assess the formed films, wear, and material transfer. It has been demonstrated that the pre-rolling process leads to the formation of a very thin compacted film with surface protective properties. With the ball as a counterpart, the graphene platelets generate a transfer film on the contacting surface.

The extent and depth of burn injury may mandate temporary use of cadaver skin (allograft) to protect the wound and allow the formation of granulation tissue while split-thickness skin grafts (STSGs) are serially harvested from the same donor areas. However, allografts are not always available and have a high cost, hence the interest in identifying more economical, readily available products that serve the same function. This study evaluated intact fish skin graft (IFSG) as a temporary cover to prepare the wound bed for STSG application. Thirty-six full-thickness (FT) 5 × 5 cm burn wounds were created on the dorsum of six anesthetized Yorkshire pigs on day −1. To mimic the two-stage clinical situation, on day 0, wounds were excised down to a bleeding wound bed and a temporary cover (either IFSG or cadaver porcine skin) was applied; then, on day 7, wounds were debrided to a viable wound bed prior to the application of autologous 1.5:1 meshed STSG (mSTSG). Rechecks were performed on days 14, 21, 28, 45, and 60 with digital images, non-invasive measurements, and punch biopsies. The IFSG created a granulated wound bed receptive to the application of an mSTSG. FT burn wounds treated with an IFSG had similar outcome measures, including contraction rates, trans-epidermal water loss (TEWL) measurements, hydration, and blood perfusion levels, compared to cadaver skin-treated burn wounds. Pathology scoring indicated significant differences between the allograft- and IFSG-treated wounds on day 7, with the IFSG having increased angiogenesis, granulation tissue formation, and immune cells. Pathology scoring indicated no significant differences once mSTSGs were applied to wounds. The IFSG performed as well as cadaver skin as a temporary cover and was not inferior to the standard of care, suggesting the potential to transition IFSGs into clinical use for burns.

Magnetic nanoparticles (MNPs), either paramagnetic or superparamagnetic depending on their composition and size, have been thoroughly studied as magnetic resonance imaging (MRI) contrast agents using in vitro and in vivo biomedical preclinical studies, while some are clinically used. Their magnetic properties responsible in some cases for high magnetization values, together with large surface area-to-volume ratios and the possibility of surface functionalization, have been used in MRI-based diagnostic and theranostics applications. MNPs are usually used as positive (T1) or negative (T2) MRI contrast agents, causing brightening or darkening of selected regions in MRI images, respectively. This review focusses on recent developments and optimization of MNPs containing Gd, Mn, Fe and other lanthanide ions which may function as dual-mode T1–T2 MRI contrast agents (DMCAs). They induce positive or negative contrast in the same MRI scanner upon changing its operational mode between T1-weighted and T2-weighted pulse sequences. The type of contrast they induce depends critically on their r2/r1 relaxivity ratio, which for DMCAs should be in the 2–10 range of values. After briefly discussing the basic principles of paramagnetic relaxation in MNPs, in this review, the basic strategies for the rational design of DMCAs are presented and typical examples are discussed, including in vivo preclinical applications: (1) the use of NPs with a single type of contrast material, Gd- or Mn-based NPs or superparamagnetic NPs with appropriate size and magnetization to provide T2 and T1 contrast; and (2) inclusion of both types of T1 and T2 contrast materials in the same nanoplatform by changing their relative positions.

Glioblastoma is the most prevalent and fatal form of primary brain tumors. New targeted therapeutic strategies for this type of tumor are imperative given the dire prognosis for glioblastoma patients and the poor results of current multimodal therapy. Previously reported drawbacks of antibody-based therapeutics include the inability to translocate across the blood–brain barrier and reach intracellular targets due to their molecular weight. These disadvantages translate into poor target neutralization and cancer maintenance. Unlike conventional antibodies, vNARs can permeate tissues and recognize conformational or cryptic epitopes due to their stability, CDR3 amino acid sequence, and smaller molecular weight. Thus, vNARs represent a potential antibody format to use as intrabodies or soluble immunocarriers. This review comprehensively summarizes key intracellular pathways in glioblastoma cells that induce proliferation, progression, and cancer survival to determine a new potential targeted glioblastoma therapy based on previously reported vNARs. The results seek to support the next application of vNARs as single-domain antibody drug-conjugated therapies, which could overcome the disadvantages of conventional monoclonal antibodies and provide an innovative approach for glioblastoma treatment.

Empathy is a multifaceted concept that is vital to effective social functioning; yet, it is impaired in high schizotypy groups. Furthermore, empathy has been found to be a mediator in the relationship between schizotypy and social functioning, highlighting the importance of empathy as a driver in social outcomes. Despite this, the four-factor structure of a widely-used measure of empathy—the Interpersonal Reactivity Index (IRI)—has been found to be psychometrically weak in high schizotypy samples. As such, this study aimed to assess differences in the item-level network of the IRI between high (n = 427) and low schizotypy groups (n = 470). The results reveal that there are significant differences in the structure of these networks, though they evidence similar strengths. Within the high schizotypy group, the network structure was consistent with the four-factor structure of the IRI subscales; items from each subscale clustered together and were distinct from those in the other subscales. By contrast, the low schizotypy group evidenced six clusters that did not mirror the IRI subscales. These results suggest that the item-level structure of the IRI is dependent upon the level of schizotypy of the sample, with the high schizotypy group’s network functioning similarly to what would be expected from the original four-factor structure.

The mechanical characteristics of graphene ribbons with an attached proof mass that can be used as NEMS transducers have been minimally studied, which hinders the development of graphene-based NEMS devices. Here, we simulated the mechanical characteristics of graphene ribbons with an attached proof mass using the finite element method. We studied the impact of force, residual stress, and geometrical size on displacement, strain, resonant frequency, and fracture strength of graphene ribbons with an attached proof mass. The results show that the increase of width and thickness of graphene ribbons would result in a decrease of the displacement and strain but also an increase of resonant frequency. The increase of the length of graphene ribbons has an insignificant impact on the strain, but it could increase the displacement and decrease the resonant frequency. The increase of residual stress in the graphene ribbons decreases its strain and displacement. The estimated fracture strength of graphene shows limited dependence on its thickness, with an estimated value of around 148 GPa. These findings contribute to the understanding of the mechanical characteristics of graphene ribbons with an attached proof mass and lay the solid foundation for the design and manufacture of high-performance graphene-based NEMS devices such as accelerometers.

Background: The greatest risk for the occurrence of the placenta accreta spectrum (PAS) is represented by uterine scars, which most frequently result after cesarean sections. Uterine rupture is a rare condition and appears mainly in the third trimester of pregnancy. The association between these two conditions is extremely rare in the first trimester of pregnancy. Methods: We performed a systematic review of abnormal placental adhesions and spontaneous uterine ruptures in the first trimester of pregnancy. We also reported a case of spontaneous uterine rupture in a 12-week pregnancy that presented with massive hemoperitoneum and hemorrhagic shock. Results: A 33-year-old patient with two previous cesarean sections, at the twelfth week of pregnancy at the time to this visit to the emergency room, presented with syncope and intense pelvic–abdominal pain. A clinical examination and ultrasound scan established the diagnosis of hemoperitoneum and hemorrhagic shock. Surgical exploration was performed, uterine rupture was identified, and hemostasis hysterectomy was necessary. The histopathological results showed placenta percreta. There have been eight reported cases of spontaneous uterine rupture in the first trimester of pregnancy associated with PAS. In these cases, it was found that 62.5% of the patients had undergone at least one cesarean section in the past; in 75% of the cases, hysterectomy was performed; and, in 87.5% of the cases, the presence of placenta percreta was confirmed. Conclusions: A high rate of cesarean sections determines the increase in the incidence of placenta accreta spectrum disorders. The possible life-threatening complications caused by this pathology can be observed in early pregnancies.

Composing coherent and structured music is one of the main challenges in symbolic music generation. Our research aims to propose a user-centric framework design that promotes a collaborative environment between users and knowledge agents. The primary objective is to improve the music creation process by actively involving users who provide qualitative feedback and emotional assessments. The proposed framework design constructs an abstract format in which a musical piece is represented as a sequence of musical samples. It consists of multiple agents that embody the dynamics of musical creation, emphasizing user-driven creativity and control. This user-centric approach can benefit individuals with different musical backgrounds, encouraging creative exploration and autonomy in personalized, adaptive environments. To guide the design of this framework, we investigate several key research questions, including the optimal balance between system autonomy and user involvement, the extraction of rhythmic and melodic features through musical sampling, and the effectiveness of topological and hierarchical data representations. Our discussion will highlight the different aspects of the framework in relation to the research questions, expected outcomes, and its potential effectiveness in achieving objectives. Through establishing a theoretical foundation and addressing the research questions, this work has laid the groundwork for future empirical studies to validate the framework and its potential in symbolic music generation.

Assuring home care (HC) workers’ safety is challenging because the work environment is a private home. This paper presents the process evaluation for a proof-of-concept safety intervention study to assess whether nurse-led safety coaching, using motivational interviewing and a safety handbook, could enable HC clients to improve safety in their homes. The process evaluation objectives were to (i) document the intervention’s implementation progress and (ii) assess the intervention’s dose delivery, dose reception, and fidelity. Five agencies employing liaisons (n = 5) and nurse managers (NMs, n = 8) implemented this study’s intervention and control arms. NMs assigned to the intervention arm (n = 6) coached 34 clients. Process evaluation metrics were assessed with mixed-methods data from (i) surveys completed by NMs during the intervention, (ii) postintervention audio-recorded and transcribed interviews (n = 6) with NMs and liaisons, and (iii) study progress tracking tools. The delivered dose efficiency was 85%, measured by the distribution of safety handbook copies to clients. About 94% of clients (n = 32) were considered “engaged” or “maybe engaged” during the safety coaching. Most coached clients (n = 30) were reachable for follow-up by NMs to assess intervention progress. Despite challenges, the intervention was implemented with good fidelity. Safety coaching can be applied in many HC contexts in larger populations.

A proof-of-concept of a microwave imaging system for the fast detection of abdominal aortic aneurysms is shown. This experimental technology seeks to overcome the factors hampering the fast screening for these aneurysms with the usual equipment, such as high cost, long-time operation or hazardous exposure to chemical substances. The hardware system is composed of 16 twin antennas mastered by a microcontroller through a switching network, which connects the antennas to the measurement instrument for sequential measurement. The software system is run by a computer, mastering the whole system, automatizing the measurement process and running the signal processing and medical image generation algorithms. Two image generation algorithms are tested: Delay-and-Sum (DAS) and Improved Delay-and-Sum (IDAS). Own-modified versions of these algorithms adapted to the requirements of our system are proposed. The system is carefully calibrated and fine-tuned with known objects placed at known distances. An experimental proof-of-concept is shown with a human torso phantom, including an aorta phantom and an aneurysm phantom placed in different positions. The results show good imaging capabilities with the potential for detecting and locating possible abdominal aortic aneurysms and reporting acceptable errors.

Colorectal cancer is a global malignancy with a high incidence and mortality rate. THZ2, a small inhibitor targeted CDK7, could inhibit multiple human tumor growths including small cell lung cancer, triple-negative breast cancer, ovarian cancer. However, the effect of THZ2 on inflammation, especially on colitis-associated colorectal cancer, is still unknown. In this study, we assessed the anti-inflammatory and anti-tumor effect of THZ2 in the mouse models of dextran sulfate sodium (DSS)-induced acute colitis and azoxymethane (AOM)/DSS-induced colitis-associated colorectal cancer. We found that THZ2 ameliorated inflammatory symptoms, including bleeding and diarrhea, in mouse models of DSS-induced acute colitis and AOM/DSS-induced colorectal cancer. The results of Western blot and immunohistochemistry showed that THZ2 rescued the up-regulated expression of COX2, IL-6, β-catenin, and snail in the mouse models. Moreover, THZ2 inhibits the development of colorectal cancer in the mouse model of AOM/DSS-induced colitis-associated colorectal cancer. Generally, THZ2 not only can inhibit DSS-induced colitis, but also can hinder AOM/DSS-induced colorectal cancer.

The formation of biogenic amines (BAs) in artisan Galotyri PDO cheeses fermented with Sterptococcus thermophilus ST1 and the Greek indigenous nisin A-producing Lactococcus lactis spp. cremoris M78 (A1cheese), or with the A1 starter supplemented with either the enterocin A-B-P-producing Enterococcus faecium KE82 (A2cheese) or the multi-functional Lactiplantibacillus plantarum H25 (A4cheese) adjunct strains was evaluated. Three pilot-scale cheese trials, GL1, GL2, and GL3, made from boiled ewes’ milk, were analyzed for their BA contents before and after cold ripening at 4 °C for 30 days. Total BAs of the fresh GL1 and GL3 cheeses (pH 4.3–4.5) were below 50 mg/kg, except for the A1/GL1 and A1/GL3 cheeses, which contained ca. 300 mg/kg (81.2% histamine) and 1250 mg/kg (45.6% putrescine) BAs, respectively. Whereas due to an outgrowth (>7 log cfu/g) of post-thermal Gram-negative bacteria contaminants during fermentation, most fresh GL2 cheeses (pH 4.7–5.0) accumulated more than 1500 mg/kg of total BAs, which exceeded 3800 mg/kg in all GL2 cold-ripened cheeses due to major increases in cadaverine and putrescine. Tyramine and histamine exceeded 500 mg/kg in the fresh A1/GL2cheeses. Conversely, total BAs remained or declined below 50 mg/kg in all cold-ripened GL3 cheeses. None of the starter or adjunct cultures could be correlated with a specific BA increase, despite E. faecium KE82, which increased at 7.6–9.2 log cfu/g in the A2 cheeses is a strong tyramine producer in culture BA broth with 1% tyrosine in vitro. The adoption of strict hygienic measures during artisan Galotyri PDO cheese production (trial GL3) enabled the best performance of all starter LAB strain combinations and reduced BA formation, whereas the high presence of Gram-negative decarboxylating bacteria contaminants compromised cheese (trial GL2) safety.
